M.U.T.H.E.R is an old super computer with a female AI created by Jonas Venture Sr. to help run The Venture Compound and act as the overseer of an underground nuclear fallout zone and make them fully automated. Her main duty also consisted of releasing small amounts of mood altering gas into the fallout zone to ensure the happiness of its future inhabitants. However, she eventually became corrupt, causing her to act irrational, possessive and make dangerously violent decisions.

Many years ago, after her mental stability began to deteriorate even worse she released a dangerous amount of hallucination-inducing gas into the fallout zone while Jonas Venture was giving a tour of it to some orphans, forcing Venture and his team to flee and leave the children behind. Team Venture and Jonas were able to shut M.U.T.H.E.R. down permanently and stop the gas before it could spread, however in their haste they were forced to seal up the fallout zone and M.U.T.H.E.R.'s core, sealing the children and fellow Team Venture member Paul Entmann inside for many years to come. Many years later, she was accidentally revived by the Venture Brothers and tried to cause World War III by launching an old nuclear missile unless her demand to see Jonas again was met. However, the missile malfunctioned due to having been filled with human wastes by the now-grown up orphans as a way to "make them clean again." Brock Samson and Entmann placated M.U.T.H.E.R. with a video reel of Dr. Jonas Venture.
