As the film begins, vampires are operating out a blood bank, luring unsuspecting donors in and killing them. A shaky, sickly looking hooded man arrives and appears ready to give blood, when the vampires reveal themselves and try to hold him down. Suddenly, the man shrieks "I hate vampires!" and attacks them. His three-way jaw opens to reveal a leech-like sucker within his mouth. The man then kills the vampires. Next, we see Blade attacking one of the vampire blood banks and fighting through a gang of vampires, killing all but one, a cowardly man named Rush. Blade leaves Rush alive, but tells him that he will come back for him. Upon return to the hideout, Blade finds his lair invaded by vampires. He, Whistler and Scud try to fight them off, only for the vampires to reveal they have not come as hostiles. Instead, they are looking for Blade's help. The trio are then taken to meet with the vampires and learn of a new crisis arising. A more devloped strain of vampirism, dubbed the Reaper virus, is sweeping through the community. The first Reaper and carrier is Jared Nomak, who was the man who attacked the blood bank at the beginning. Eli Damaskinos, the vampire overlord, has enlisted Blade to help him combat the new threat. Blade is introduced to Damaskinos and his human familiar, Karl Kounen. Blade then agrees to go along with the uneasy alliance. Blade is teamed with the Bloodpack, a team of vampires who were originally assembled to kill Blade. The team is led by Damaskinos' daughter Nyssa and consists of Reinhardt, Chupa, Asad, Snowman, Priest, Lighthammer and his girlfriend Verlaine. Right away, there is some tension between them, most notably between Blade and Reinhardt. The group begins by heading to a vampire nightclub. Outside, Whistler covers the team as a sniper while Scud sits in the transport. The mission goes awry when the Reapers appear but turn out to be immune to immune to silver and garlic. A gang of Reapers attack the transport, but Scud is able to kill them by using a massive UV powered light. At the same time, Whistler vanishes. In the club, a Reaper attacks Priest and badly injures him, while another bites Lighthammer, who hides the wound from the others. Blade and Nyssa encounter Nomak, who for some reason doesn't kill Nyssa. Blade and Nomak fight, the battle only ending when Nomak must flee from the sunlight. In the aftermath, Priest begins to change into a reaper, forcing Blade to kill him using sunlight. Suddenly, Whistler reappears, revealing that he discovered a dying Reaper at the entrance to the sewer, revealing the location of the Reaper's nest. While Whistler and Scud build UV weapons for the team, the others disect the dead Reaper and find that the Reaper's hearts are encased in bone, making them even more difficult to kill. Blade prepares the team for an invasion of the Reaper nest. Once inside of the sewers, Lighthammer succumbs and changes. He corners and kills the unsuspecting Snowman, and then Verlaine. Verlaine climbs up a ladder and opens a sewer hatch, killing both of them as they are drenched in sunlight. Meanwhile, Chupa decides to retaliate for Priest's death by killing Whistler, attempting to beat him to death. Whistler, however, is able to spray Chupa with Reaper pheromones and leaves him to die at the hands of the Reaper horde. Nyssa and Asad locate the center of the nest, when suddenly Reapers burst out of the water and drag Asad beneath, killing him. Blade arrives in time to save Nyssa. Blade then uses a cluster UV device to wipe out all of the Reapers, but the light also scars Reinhardt's face. Whistler encounters Nomak, who gives him a ring to give to Blade, and lets him go. Suddenly, gangs of armed vampire familiars flood into the sewers and capture Blade and Whistler. Scud tries to rescue them, but is killed by Reinhardt. At Damaskinos' stronghold, Whistler gives Blade the ring, who examines it and realises what Nomak wants. Damaskinos enters and Blade shows him the ring, which is the ring of the Damaskinos family. Damaskinos explains that he hoped to create a legacy of perfect vampires, and designed the Reaper virus. His son, Nomak, was the first. Nomak began attacking the vampires as he wanted revenge. Damaskinos then has Blade taken to be disected, in order to perfect the strain and create more daywalkers. Whistler manages to escape and climb through a ventilation shaft, stealing a gun and shooting Kounen to death, before rescuing Blade. Blade then fights through Damaskinos' men, saving Reinhardt to last. Blade manages to cut Reinhardt in two, killing him. Nomak arrives at the base and breaks in, slaughtering his father's men. Damaskinos attempts to escape to a helicopter, but Nyssa, disillusioned by his extreme methods, cuts him off and locks down the building, trapping her father inside with Nomak. Damaskinos tries to show Nomak that he loves him and is willing to help, but Nomak sees through his lies and brutally murders him. Then, to complete the circle, he bites Nyssa. Blade enters and he and Nomak engage in a final showdown. Blade pinpoints Nomak's weak point and stabs him under his arm, bypassing the bone which encases his heart. Blade then kicks the sword deeper into Nomak, sending it straight into the villain's heart. Nomak thanks Blade for freeing him before dying. Blade rushes to Nyssa's side, but she tells him that it's too late. Knowing that she will soon change, Nyssa requests that she be allowed to die as a vampire, how she was born. She then asks to finally see the sunrise. Blade carries her outside as the sun comes up and she disintergrates in his arms. At the end of the film, with the crisis over and the vampire community thrown into turmoil due to Damaskino's death, Blade pays Rush a visit in a shady strip club, asking him if he had thought he forgot about him.
