- The U-3 is one of the experiments carried out by the research team at the Island. It is a horrifying amalgamation of many beings all fused imperfectly into one. While it has the upper part of a human torso and a human head, its visage is monstrously warped and marked by strange scars upon its nape and crown, a clawed arm, and a tentacle in place of the other one. Below this, it is grafted in a centaur-like fashion into a deformed body, that has two front legs and two hind ones bonded by a centipede-like fleshy piece of muscle. Later, after receiving enough damage, the Plaga attached to its spine emerges to attack. The Plaga has evolved since its first stages and resembles a tentacle ending in a gaping mouth, lined with dozens of teeth. This tentacle also sports a heavy pair of pincers.

- Le U-3 possède les traits physiques caractéristiques de chacun des animaux qui composent son ADN complexe. Son aspect vulgaire et son instabilité biologique dénote un certain amateurisme de la part des chercheurs qui l'ont créé. Son torse est humain, tout comme son visage, arborant une grimace atroce. Des cicatrices sont visibles sur sa nuque et le sommet de son crâne. Son bras droit est pourvu de griffes et son bras gauche est un tentacule. Il utilise tous deux pour attaquer ses adversaires. La partie postérieure de son corps rappelle un scolopendre.

| - L'U3 (Chiamato Lui da Saddler) è una creatura formata dall'unione delle cellule di un rettile, di un insetto e di un umano in un unico organismo. Incredibilmente agile e veloce, possiede una Plaga con delle tenaglie nella bocca, capace di decapitare il nemico, che emerge direttamente dal suo corpo quando esso è gravemente danneggiato. Nella prima parte dell'icontro, che avviene nei Containers, è impossibile ucciderlo, ma è possibile stordirlo e farlo fuggire con le armi. Nella seconda parte dello scontro, che avviene su una piattaforma rocciosa, bisognerà sparargli con tutte le armi a disposizione, sfruttando di tanto in tanto i barili di esplosivo disseminati per il campo. U3 sfrutta anche degli attacchi a sorpresa scavando sottoterra e sbucando all'improvviso.
