In the year 1968, every day is the same for Dr. Robert Morgan: he wakes up, gathers his weapons and then goes hunting for vampires.   • In the year 2068, a virus wipes out the entire male population of the planet. A narrator (voiced by Jeff Goldblum) explains that in this near future women have left the house completely. They no longer feel the need for men and traditional house wife duties, its become so bad women no longer even learn how to cook and thus order out every night or force their good-natured husbands to cook the meals for them while they go off and run companies in their pant suits. Due to this total lack of care for womanly duties when the virus hits killing all men they are unable to provide for themselves, after cutting themselves off for so long from men. The all female population them becomes split on what to do. Some believe that they should start taking on the roles of men while others believe they should use science to create a new population of men and start over. As they continue to argue a war breaks out between the two sides. Unbeknown to the feuding women there is still one lone man alive on earth his name is Robert Omega (Crispin Glover). Robert for a reason never explained has survived the virus attack, but his fear of women keeps him from revealing his secret to the world. He spends his days hiding out in an abandoned Hooters Restaurant. Eventually he is discovered by a anti-feminist woman played by Amy Adams named Regina. Regina and Robert fall in love and then in an unexplained 15 minute overly graphic sex scene also map out a plan to escape to the one place not at war, Antarctica. Trouble is brewing however, as the Queen of New Europe Ellen (Lucy Pinder) is gaining in power and has already taken over Asia. Ellen plans to take over the entire planet and banish science as she always hated men and sees this as an opportunity to guarantee they never exist. Her only true opposition is General Tanya Smith (Jennifer Aniston) who alongside Admiral Beyonce have been pushing back her attempts at expanding her empire. Violence ensues as Regina and Robert disguise themselves as sailors to escape to Antarctica, but end up in a large naval battle on the south coast of Africa. Beyonce is killed in the battle when she falls overboard and instead of trying to swim tries to protect her weave. While Beyonce dies her weave does manage to survive but decides to drown itself rather than live on without its master. This is often described as one of the more touching moments in the film. Regina and Robert eventually make it to Antarctica but do not have proper clothing for the weather. Minutes from freezing to death they are rescued by a researcher played by Gilbert Gottfried named Sam. Sam takes the two to his lab and there for no reason at all attempts for a threesome. Once he is shot down, he explains that he created the man killing virus and that the only reason he survived was because he had the antidote. Regina is enraged and kills Sam with a pistol, by clubbing him to death. Robert and Regina embrace each other and decide to repopulate the world together, but before they can the lab is blown up by a rogue missile fired by Tanya. Regina and Robert are killed in the explosion. Later Tanya searches the rubble of the lab and realizes that her army has killed the last man on earth. Distraught she surrenders to Ellen and signs a treaty giving the world over to Ellen's empire. After signing the treaty there is a massive lesbian orgy featuring both armies. The final image is of Tanya Smith dancing drunk on the beach naked.

A flashback sequence explains that, three years before, Morgan's wife and daughter had succumbed to the plague, before it was widely known by the public that the dead would return to life. Instead of taking his wife to the same public burn pit used to dispose of his daughter's corpse, Morgan buried her without the knowledge of the authorities. When his wife returned to his home and attacked him, Morgan became aware of the need to kill the plague victims with a wooden stake. Morgan hypothesizes that he is immune to the bacteria because he was bitten by an infected vampire bat when he was stationed in Panama, which introduced a diluted form of the plague into his blood. One day, a dog appears in the neighborhood. Morgan chases after the dog but does not catch it. Some time later, the dog appears, wounded, at Morgan's doorstep. He takes the dog into his home and treats its wounds, looking forward to having company for the first time in three years. He quickly discovers, however, that it too has become infected with the plague. Morgan is later seen burying the dog, which he has impaled with a wooden stake. While out on his daily rounds, Morgan spots a woman in the distance. The woman, [[Ruth[[, is terrified of Morgan at first sight, and runs from him. Morgan convinces her to return to his home, but is suspicious of her true nature. Ruth becomes ill when Morgan waves garlic in her face, but claims that she has a weak stomach. Morgan's suspicion that Ruth is infected is confirmed when he discovers her attempting to inject herself with a combination of blood and vaccine that holds the disease at bay. Ruth initially draws a gun on Morgan, but surrenders it to him. Ruth then tells him that she is part of a group of people like her — infected but under treatment — and was sent to spy on Morgan. The vaccine allows the people to function normally with the drug in the bloodstream, but once it wears off, the infection takes over the body again. Ruth explains that her people are planning to rebuild society as they destroy the remaining vampires, and that many of the vampires Morgan killed were technically still alive. While Ruth is asleep, Morgan transfuses his own blood into her. She is immediately cured, and Morgan sees hope that, together, they can cure the rest of her people. Moments later, however, Ruth's people attack. Morgan takes the gun and flees his home while the attackers kill the vampires gathered around Morgan's home. Ruth's people spot Morgan and chase him. He exchanges gunfire with them, and picks up tear gas grenades from a police station armory along the way. While the tear gas delays his pursuers somewhat, Morgan is wounded by gunfire and retreats into a church. Despite Ruth's protests to let Morgan live, his pursuers finally impale him on the altar with a spear. With his dying breaths, Morgan denounces his pursuers as "freaks," and declares that he is the last true man on earth.
