After you have written a quest (in the Quest Editor) and made an asset for the quest file (in the Art Manager), you must add the Quest to your map. This is done here. After choosing 'Add Quest' from the menu, click 'add' in the dialog window. From the list of quests that appears, select all quests you want to use on your map (by shift-clicking) After the next build the quests will be active.
