It's not the Polyjuice Potion, but the Draught of Living Death, where clockwise stirs come into play. During a Potions lesson in Half-Blood Prince, Harry is brewing the Draught of Living Death, and one of the handwritten notes in his Advanced Potion-Making book says to add one clockwise stir after every seven counterclockwise stirs to finish the Draught faster.
