After seeing Giacomo at the entrance of of Rodolfo's Estate, the Tree-hugging Granny can be found standing across from the pub. Agree to help her move the bugs out of the way in Nunki Valley and she will give you the Holy Leaf, which will allow you to carry the Holy Droplets necessary for the quest. Return to her after you've moved all of the bugs.
