Calvin Zabo created this drug after his daughter, Daisy Johnson, was taken from him by S.H.I.E.L.D. Cal was convinced that if he had been stronger, he could've protected Daisy and, after being tortured by Dr. Whitehall, Jiaying had no interest in ordinary humans, thus Cal created this drug to give himself the power he wasn't born with.
