The show follows the lives of Mario, Woody, Shrek, Bowser, Bowser Jr., Chef Pee Pee, Mama Luigi, and all the other SML Movie characters in their lives. The show diverges from the YouTube series in that the characters live in a mansion called the Mark H. Stephens Mansion (using the Munster Mansion set at Universal Studios) rather than in separate houses. The mansion's landlord is Lisa von Babbel, a Dutch immigrant (played by Anneliese van der Pol), and is the only human who can talk to the plushies (though other people dismiss it as insanity).
