I would say only save images as PNG before applying transparency. BMP isn't compatible with the wiki, and GIF should be used when creating animated images only (yes I know PNG can also do animated, but an animated PNG isn't compatible with many browsers). Your next best bet in terms of quality is SVG, which wouldn't make sense to use anyways, since the vast majority of Nitrome's games don't use vector graphics.
